Introduction

Microbial community analysis based on small-subunit rRNA genes as well as protein-coding gene clone libraries has become commonplace in microbial ecology. Comparison and characterization of many microbial samples in terms of the phylogeny of the microbes are particularly valuable for factors that affect microbial distribution. There already have some programs for OTU (operational taxonomic unit) assignment and microbial identification. But up to now, there have no programs suitable for self-dataset construction and update, and for the analysis of other genes including 18S rRNA gene.

Program Dscription and Application

OTU_Misess is an integrated tool for OTU assignment and Microbial Identification of Sequences from Environmental SampleS. The input of OTU_Misess are chromatograph (or fasta format) files sequenced from cloned samples collected in various environments. OTU_Misess can be used to construct and update a local OTU database and assign taxonomy information from public domain databases to local OTUs. Additional information comprising the terminal restriction fragment (T-RF) length of each sequence is useful for terminal restriction fragment length polymorphism (T-RFLP) analysis. OTU_Misess is not only useful for 16S rRNA gene analysis but also useful for 18S rRNA gene, ITS (internal transcribed spacer) or any other functional genes. On our website, taxonomical databases of 16S rRNA, 18S rRNA gene (include ITS) are provided and updated frequently for user convenience.
